Certain actin-related proteins (Arps) of budding yeast are localized in the nucleus, and have essential roles as stoichiometric components of histone acetyltransferase (HAT) and chromatin remodeling complexes. On the other hand, identification of vertebrate nuclear Arps and their functional analyses are just beginning. We show that human Arp5 (hArp5) proteins are localized in the nucleus, and that arp5Δ yeast cells are partially complemented by hArp5. Thus, hArp5 is a novel member of the nuclear Arps of vertebrates, which possess evolutionarily conserved functions from yeast to humans. We show here that hArp5 shuttles between the nucleus and the cytoplasm. Furthermore, after the induction of DNA double strand breaks (DSB), cell growth and the accumulation of phosphorylated histone H2AX (γ-H2AX) are impaired by hArp5 depletion. Association of hArp5 with the hIno80 chromatin remodeling enzyme and decrease of chromatin-bound hIno80 by hArp5-depletion indicate that hArp5 may have a role in the recruitment of the hINO80 complex to chromatin. Overexpression of hArp5 and hIno80 enhanced γ-H2AX accumulation. We determined the fruit predators and seed dispersers of two species of durian ( Durio , Bombacaceae), D. graveolens and D. zibethinus , which have contrasting aril color and timing of dehiscence by observing fruiting trees in Deramakot forest reserve. In our observations in the wild, both species were extensively predated by orangutans before fruits matured. Durio graveolens was dispersed by black hornbills and D. zibethinus was dispersed by long-tailed macaques. 相似文献

Structural elucidation of an intensely blue fluorescent compound (A) formed from sepiapterin by Bacillus subtilis is described. The structure of the catabolite (A) was found to be 2-amino-6-(1-carboxyethoxy)-4(3H)-pteridinone (9) from both spectroscopic and degradation studies. This was confirmed by an unambiguous synthesis of 9. The stereochemical structure of the side chain at the 6-position of A was confirmed to be the L(or S) configuration, as in sepiapterin, by analysis of the lactic acid formed from A on acid hydrolysis. This suggests that the side chain is rearranged intact during the catabolic conversion of sepiapterin. A possible mechanism for the conversion is discussed. 相似文献

Summary In a field experiment soil samples buried at the warmer temperature regime nitrified added ammonium faster than soils buried at the cooler temperature regime. Nitrification occurred more rapidly under both regimes in a soil which had developed in a warm climatic zone than in two other soils developed under cooler conditions.The rate of nitrification of added ammonium in soils incubated at 5, 15, 25 and 40°C in the laboratory increased with increase in the temperature up to 25°C in three out of four soils. In the fourth soil nitrification was as active at 40°C as at 25°C. The temperature range for appreciable nitrification to occur in a soil was related to the environmental conditions where the soil was formed.Mineralization of organic nitrogen occurred to a greater extent at 40°C than at three lower incubating temperatures of 5, 15, and 25°C. Immunoblotting analysis revealed heterologous distribution profiles of the non-N-methyl-D-aspartate (NMDA) receptor subunits, GluR1, GluR2 and GluR6, in membrane fractions prepared from murine discrete brain structures including hippocampus. In coronal sections fixed with paraformaldehyde (PA) solution after dissection from mice perfused with 4% PA, however, no marked immunoreactivity was detected to GluR6 subunit in any hippocampal subregions, with high immunoreactivities to both GluR1 and GluR2 subunits in the strata oriens, radiatum and lacunosum-moleculare of the CA1 and CA3 subfields and the stratum moleculare of the dentate gyrus in hippocampus. In coronal, sagittal and horizontal sections fixed with Carnoy solution after dissection from animals decapitated, by contrast, high immunoreactivity was additionally detected to GluR6 subunit in the stratum lucidum of hippocampus. The systemic administration of kainate not only resulted in marked neuronal losses along the CA1-CA4 pyramidal layers 1 week later, but also led to significant decreases in immunoreactivities to GluR1, GluR2 and GluR6 subunits in the CA1 and CA3 subfields on brain coronal sections prepared by immersion fixation with Carnoy solution. These results suggest that immersion fixation with Carnoy solution may be suitable and appropriate for reproducible and quantitative immunohistochemical detection of particular non-NMDA receptor subunits in murine hippocampus. 相似文献

Molecular phylogenetic analyses of representative Cutleria species using mitochondrial cox3, chloroplast psaA, psbA and rbcL gene sequences showed that C. cylindrica Okamura was not included in the clade composed of other Cutleria species including the generitype C. multifida (Turner) Greville and the related taxon Zanardinia typus (Nardo) P.C. Silva. Instead, C. cylindrica was sister to the clade composed of the two genera excluding C. cylindrica. Cutleria spp. have heteromophic life histories and their gametophytes are rather diverse in gross morphology, from compressed or cylindrical‐branched to fan‐shaped, whereas the sporophytes are rather similar. In contrast, the monotypic species Z. typus has an isomorphic life history and resembles fan‐shaped Cutleria in morphology. Morphological comparisons of these taxa revealed that C. cylindrica is morphologically distinct from other Cutleria spp. and Z. typus in having cylindrical gametophytes with multiseriate trichothallic filaments instead of uniseriate filaments (hairs) characteristic of Cutleriales (or Cutleriaceae, Tilopteridales), and in lacking rhizoidal filaments in the crustose sporophytes. Therefore, based on the molecular and morphological data, the establishment of a new genus Mutimo to accommodate C. cylindrica, and the new combination of M. cylindricus, is proposed. 相似文献

The paranodal junction is a specialized axon-glia contact zone that is important for normal neuronal activity and behavioral locomotor function in the central nervous system (CNS). Histological examination has been the only method for detecting pathological paranodal junction conditions. Recently, diffusion tensor MRI (DTI) has been used to detect microstructural changes in various CNS diseases. This study was conducted to determine whether MRI and DTI could detect structural changes in the paranodal junctions of the spinal cord in cerebroside sulfotransferase knock-out (CST-KO) mice. Here, we showed that high-resolution MRI and DTI characteristics can reflect paranodal junction failure in CST-KO mice. We found significantly lower T1 times and significantly higher T2 times in the spinal cord MRIs of CST-KO mice as compared to wild-type (WT) mice. Spinal cord DTI showed significantly lower axial diffusivity and significantly higher radial diffusivity in CST-KO mice as compared to WT mice. In contrast, the histological differences in the paranodal junctions of WT and CST-KO mice were so subtle that electron microscopy or immunohistological analyses were necessary to detect them. We also measured gait disturbance in the CST-KO mice, and determined the conduction latency by electrophysiology. These findings demonstrate the potential of using MRI and DTI to evaluate white matter disorders that involve paranodal junction failure. 相似文献
